I feel what really defines it is the gameplay style. While it has almost always been associated with horror themes, save for Squawkers, it's really the whole PMD combined with mystery and puzzle solving that sets it apart from everything else out there.
I mean heck, Xanthe was able to map out and create an entire season all on her own. And she had a unique style to it and still it totally worked great, so it's a very adaptable system that works with different styles. I also like the fact it feels a bit more open world and not very linear.
